Can Daku no Oni use his ability to destroy an opponent farm holding while he's unopposed?
Yes.
The required targets of the action are "a Personality opposing Daku" or "a Holding controlled by the enemy leader", (not both), so you can pick either. The Attacking or Defending player is always the "enemy leader" from the other's perspective, whether he assigns units to oppose you or not, so that's covered. And the Rule of Location only restricts targeting cards in units to ones that are at the battlefield; a Holding could never be at the battlefield anyway (in SE).
So, there you have it. Nom nom nom those Farm Holdings away!
