If in a game with more than 2 players, if I have Hida Osote out, and both players target one of my personalities at his location do they both discard a card once per turn, or just the first player to target my personality

Only the first one. Osote's trigger is the first time each turn another player's action targets your Crab Clan Personality at Osote's location, not the first time per player.
