Cannot gain Corruption tokens or Shadowlands. Dynasty/Open, b: Kachiko may remain bowed. Target a Personality. While Kachiko remains bowed, after each time the target assigns, the target's controller's turn begins, and your turn begins if the target is Shadowlands, give the target a -1F/-1C Poison token. Remove these tokens if Kachiko straightens or leaves play.

So the bolded part requires the target to be shadowlands, is that a consideration for all 3 clauses for her ability?

1.) So the target when assigning, do they have to have the shadowlands keyword to get the token? 2.) Likewise, at the start of the target's turn, will they have to have the shadowlands keyword to get a token? 3.) Or is only on the Kachiko player's turn, that they will have to have the keyword in order to get a token?

No, it is not.

Kachiko has 3 independent triggers. Only the third requires the target to be Shadowlands to trigger. Your example #3 is correct.
