Now that targeting occurs before interupts, when someone targets one of my courtier at Dakatsu's location, even if I Final Sacrifice it to a Yohimbo, do they still lose the two honor for the original targeting?

No. Effects based off targeting trigger at step E of the action sequence, and at that point, the change in targets to the Yojimbo has occurred.
